Summary

Researchers investigated the effects of Patolakaturohiniyadi Kashayam (PKR), a classical Ayurvedic formulation, on the regulation of lipid metabolism and improvement of metabolic dysfunction associated with obesity. The study used an integrated approach, including network pharmacology and analyses in cellular and animal models. Results showed that PKR significantly reduced lipid accumulation and triglyceride levels in hepatic and adipose cells. In an animal model fed a high-fat and high-fructose diet, PKR decreased body, liver, and adipose tissue weight, improved blood lipid profiles, and reduced levels of liver enzymes AST and ALT. Additionally, PKR improved glucose tolerance and elevated levels of GLP-1, a hormone that helps regulate appetite and glucose. Identified mechanisms include modulation of genes such as TP53, AKT1, IL6, TNF, and STAT3, which are involved in the regulation of lipid metabolism and inflammation. These findings highlight the potential of PKR as a multi-target therapy for metabolic dysfunctions.

Key points

  • PKR reduced the accumulation of lipid droplets and intracellular triglycerides in hepatocytes and adipocytes.
  • Identification of molecular targets such as TP53, AKT1, IL6, TNF, and STAT3, related to lipid metabolism and inflammation.
  • In an animal model, PKR decreased body, liver, and adipose tissue weight, in addition to improving lipid profiles and reducing AST and ALT.
  • PKR restored phospholipid balance, increasing lysophospholipids and phosphatidylethanolamines.
  • Improvement in glucose tolerance and increased levels of GLP-1, an important hormone in appetite regulation.

✦ For you

If you are concerned about weight gain or liver health, consider observing your diet and including more foods that promote metabolic health, such as vegetables, whole grains, and lean protein sources. Additionally, regular exercise can help improve metabolism and overall health. If you notice persistent symptoms, seek a qualified healthcare professional.

For practitionersExpand

In clinical practice, consider including Patolakaturohiniyadi Kashayam (PKR) in protocols for patients with metabolic dysfunction and obesity. Evaluate the patient's condition regarding lipid metabolism and inflammation, and adjust formulas as needed. Combining PKR with dietary and lifestyle guidance may enhance results.
